Abstract

The Human Genome in Health and Disease: Therapeutic Perspectives and Genoprotective Approaches

ABSTRACT

The human genome, comprising over three billion base pairs and approximately 20,000–25,000 genes, represents the fundamental blueprint of life. It encodes the instructions for cellular processes, protein synthesis, and inheritance, thereby shaping physical traits, physiological functions, and disease susceptibility. Defects in the genome, arising from mutations, chromosomal abnormalities, or impaired DNA repair mechanisms, can lead to a wide spectrum of disorders such as cystic fibrosis, sickle cell anemia, Huntington’s disease, Down syndrome, Duchenne muscular dystrophy, and cancer. These conditions vary in severity, inheritance patterns, and clinical manifestations, often resulting in significant health and developmental challenges. Advances in genomics, including next-generation sequencing, bioinformatics, and molecular biology, have transformed the diagnosis and management of genetic disorders. Early detection through genetic testing and prenatal screening has improved clinical outcomes, while emerging therapies such as CRISPR-Cas9 gene editing and personalized medicine offer promising avenues for correcting or mitigating genomic defects. Parallel to these developments, genoprotective agents—both natural and synthetic—have gained attention for their ability to safeguard DNA integrity. Natural compounds like curcumin, resveratrol, and quercetin, along with vitamins C and E and minerals such as selenium and zinc, act as antioxidants and enhance DNA repair pathways. Synthetic agents such as amifostine provide clinical protection against genotoxic stress during chemotherapy and radiotherapy. The modes of action of genoprotective agents include free radical scavenging, DNA repair enhancement, chromatin stabilization, anti-mutagenic activity, and immune modulation. These mechanisms collectively reduce the risk of mutations, genomic instability, and disease progression. Applications span oncology, radiology, and preventive medicine, with nutraceuticals and functional foods enriched with genoprotective compounds increasingly promoted for healthy aging and disease prevention. In conclusion, the integration of genomic research with therapeutic innovations and genoprotective strategies holds immense potential for advancing healthcare. Future perspectives emphasize precision medicine, gene editing, and nutraceutical interventions as key pillars in reducing the global burden of genetic disorders. Collaborative research across genomics, pharmacology, and clinical sciences will be essential to translate these advances into accessible and effective therapies.
